Short answer, not recommended.
Here's why.
Silicon skins in a minute or two, but takes days to be fully cured, and it doesn't hold well on older silicon.
So it is usually recommended that you fully remove the old silicon, clean the area thoroughly with rubbing alcohol, then apply new silicon, after that give it a few days, I would recommend at least 1 week for it to be fully cured before adding water.
Having water in your tank also added the problem of outward pressure exerted by your tank water. It pushes the glass further apart, would be better if you dry the tank before doing your repairs.
